Complete the main quest, 'The Caretaker's Lunar Lament'
To complete the main quest, The Caretaker's Lunar Lament, you must meet with Gladwin Moon, the caretaker of Hogwarts, who will introduce you to the Demiguise Statues. You must then observe how the statue changes when day switches to night. Once it does, remove the Demiguise Moon from the statue.
Follow Gladwin to a nearby locked door, where he will ask you to deal with two additional Demiguise Statues. He will then teach you Alohomora, an essential spell that will always be equipped without needing a Spell Slot. To learn the spell, you must complete a spell-learning minigame.
Use Alohomora to open the locked door by rotating both sets of gears until you see a reaction, and then holding them in place. This will initiate a minigame that can be challenging to understand at first. Once you have unlocked the door, cast Disillusionment and proceed through.
Head upstairs and cast Revelio to reveal nearby prefects and professors, who you will want to avoid. You can use Basic Cast to lure them away. On the first floor, there will be an Arithmancy door that you can complete, but it is optional. Head up the spiral staircase to access the Prefect's Bathroom via a Level 1 locked door. Unlock it with the Alohomora minigame, then proceed around the right side of the bathroom to avoid the prefects and find a Demiguise statue.
Head back out and up the spiral staircase to your left to arrive at the Hospital Wing Floo Flames location. Sneak past the nurse and professor near one of the beds to find the second Demiguise statue on a table at the far end of the room.
Once you have both Demiguise Moons, return to Gladwin Moon to complete the quest.
